Bio-based Polymers from Epoxidized Vegetable Oils Modified by Metathesis
The epoxides of oligomeric self-metathesized soybean oil (MSBO) and cross-metathesized palm oil (PMTAG) and canola oil (CMTAG) containing terminal double bonds were used to produce nonisocyanate polyurethanes (NIPUs) as well as anhydride, amine and thiol-cured epoxies. The synthesized NIPUs displayed metal adhesive properties for CMTAG and MSBO with MSBO being favoured probably due to its plasticizing nature as opposed to CMTAG. The relationship was reversed for the anhydride curing reaction where it was found that CMTAG, due to its lower degree of plasticizing content from the reduced dangling chains, produced higher tensile strengths than MSBO. Both MSBO and CMTAG led to fully cured amine and thiol products that were sticky gels, which prevented physical analyzses of their expected solidified products. PMTAG, due to its low number of reactive groups, was not suitable for the synthesis of these polymers. Beyond Habitat
My objective was to understand how individual variation, in conjunction with variation in habitat, can affect individual and population-level variation in animal space use. I used coyotes (Canis latrans) as a model species to investigate the roles of hybridization, an inherited intrinsic factor, and spatial memory, a learned intrinsic factor, on space use. I used a diversity of methods and approaches, including meta-regression, multiple imputation, simulations, resource selection functions, step selection functions, net-squared displacement analysis, and survival analysis. A major contribution was my investigation of the performance of multiple imputation in a meta-regression framework in Chapter 2. My simulations indicated that multiple imputation performs well in estimating missing data within a meta-regression framework in most situations. In Chapter 3, I used published studies of coyote home range size in a meta-regression analysis with multiple imputation to examine the relative roles of hybridization and environmental variables on coyote home range size across North America. I found that hybridization with Canis species was a leading factor driving variation in coyote space use at a continental scale. In Chapter 4, I used telemetry data for 62 coyotes in Newfoundland, Canada, to investigate the influence of cognitive maps on resource use. I found that resident coyotes used spatial memory of the landscape to select or avoid resources at spatial scales beyond their immediate sensory perception relative to transient coyotes, presumably increasing their fitness. Taken together, my dissertation demonstrates that intrinsic factors, such as genetic ancestry and spatial memory, can have substantial influences on how animals use space at both individual and population levels, and at both a local and a continental scales. Behavioural ecology and population dynamics of freshwater turtles in a semi-urban landscape at their northern range limit
Species are faced with a variety of challenges in the environment, including natural challenges, such as variability in ambient temperature, and anthropogenic threats, such as habitat transformation associated with urbanisation. Understanding how animals respond to these kinds of challenges can advance the field of behavioural ecology and guide management decisions for wild species. Yet, we still have limited understanding of the extent of natural and human-caused impacts on animal behaviour and population dynamics, and lack robust assessment of behaviour in free-ranging animals. Using novel miniaturised biologging technologies, I characterised and validated behaviour in two freshwater turtle species: Blanding’s turtles (Emydoidea blandingii) and Painted turtles (Chrysemys picta). Further, I investigated how these two ectothermic species navigate a thermally heterogeneous landscape near their northern range limit, by comparing selected and available ambient temperatures. I showed that turtles preferred locations that were, on average, warmer and less variable in temperature than the available environment, and that this thermal sensitivity was greatest early in the year, and at fine spatial scales that likely matched the species' perception of the environment. Lastly, I assessed whether urban development was compatible with long-term viability of a Blanding’s turtle population, by monitoring habitat change and turtle survival over one decade of ongoing residential and road development. I found that Blanding’s turtle habitat quantity and connectivity declined in the area, which coincided with high road mortality and severe declines in turtle survival and population size, especially in adult females. I concluded that urban development and current road mortality rates are incompatible with the long-term viability of this at-risk turtle population. Overall, my findings demonstrate the importance of variation in the thermal environment and anthropogenic impacts on habitat in shaping the behaviour and population dynamics of this species-at-risk. Bank Swallow (Riparia riparia) Breeding in Aggregate Pits and Natural Habitats
I examined Bank Swallow (Riparia riparia) colony persistence and occupancy, in lakeshore, river and man-made aggregate pit habitat. Habitat persistence was highest on the lakeshore and lowest in aggregate pits, likely due to annual removal and relocation of aggregate resources. Bank Swallow colonies in aggregate pit sites were more likely to persist if a colony was larger or if burrows were located higher on the nesting face. I also compared nest productivity and health factors of Bank Swallows in lakeshore and aggregate pit habitats. While clutch size was the same in both habitat types, the number of fledglings from successfully hatched nests was significantly higher in aggregate pit sites than from lakeshore sites. Mass of fledgling Bank Swallows did not differ significantly between habitat types, however mass of adults from aggregate pits decreased significantly over the nesting season. Parasite loads on fledgling Bank Swallows were significantly lower in aggregate pits than in lakeshore sites. According to these indicators, aggregate pits appear to provide equivalent or higher quality habitat for Bank Swallows than the natural lakeshore sites, making them adequate and potentially key for this species’ recovery. Aggregate pit operators can manage for swallows by (1) creating longer, taller faces to attract birds and decrease predation, and (2) supplementing their habitat with water sources to encourage food availability. Automated Separation and Preconcentration of Ultra-Trace Levels of Radionuclides in Complex Matrices by Online Ion Exchange Chromatography Coupled with Inductively Coupled Plasma Mass Spectrometry (ICP-MS)
Radionuclides occur in the environment both naturally and artificially. Along with weapons testing and nuclear reactor operations, activities such as mining, fuel fabrication and fuel reprocessing are also major contributors to nuclear waste in the environment. In terms of nuclear safety, the concentration of radionuclides in nuclear waste must be monitored and reported before storage and/or discharge. Similarly, radionuclide waste from mining activities also contains radionuclides that need to be monitored. In addition, a knowledge of ongoing radionuclide concentrations is often required under certain ‘special’ conditions, for example in the area surrounding nuclear and mining operations, or when nuclear and other accidents occur. Thus, there is a huge demand for new methods that are suitable for continuously monitoring and rapidly analyzing radionuclide levels, especially in emergency situations. In this study, new automated analytical methods were successfully developed to measure ultra trace levels of single or multiple radionuclides in various environmental samples with the goal of faster analysis times and less analyst involvement while achieving detection limits suitable for typical environmental concentrations. Assessment of the impacts of noise and vessel traffic on the distribution, abundance and density of Chinese humpback dolphins (Sousa chinensis chinensis) in the waters of Hong Kong
Marine mammals with near-shore distributions are susceptible to human-related recreational and commercial disturbances, particularly near densely populated and industrialized coastal areas. A population of over 2,500 Chinese humpback dolphins (Sousa chinensis chinensis) occupies the Pearl River Estuary in southern China. A part of this population uses Hong Kong’s waters off of Lantau Island, where they are subjected to a number of anthropogenic threats, including vessel disturbance, fisheries interactions, and boat-based tourism. Previous research has shown that the abundance of this subspecies in Hong Kong’s waters has declined about 60% since 2003. Using a combination of acoustic recordings, dolphin distribution and abundance data, and vessel traffic information I found that: 1) Four types of vessels common to the waters on Hong Kong generate noise that is audible to Sousa chinensis chinensis; 2) The spatial distribution of underwater noise in Hong Kong’s waters does not significantly vary among the six sites sampled; 3) High-speed ferry traffic and passenger volume has increased dramatically during the study period; 4) There has been a significant decline in dolphin density in areas within and near vessel traffic; and 5) Dolphins are most at risk of vessel collisions and being exposed to vessel noise near Fan Lau and within the Urmston Road waterway just northeast of the Sha Chau and Lung Kwu Chau Marine Park . These results can inform future acoustic studies on this species and guide conservation and management efforts in Hong Kong. Assessment of an adult lake sturgeon translocation (Acipenser fulvescens) reintroduction effort in a fragmented river system
North American freshwater fishes are declining rapidly due to habitat fragmentation, degradation, and loss. In some cases, translocations can be used to reverse local extirpations by releasing species in suitable habitats that are no longer naturally accessible. Lake sturgeon (Acipenser fulvescens) experienced historical overharvest across their distribution, leading to endangered species listings and subsequent protection and recovery efforts. Despite harvest and habitat protections, many populations do not appear to be recovering, which has been attributed to habitat alteration and fragmentation by dams. In 2002, 51 adult lake sturgeon from the Mattagami River, Ontario, Canada were translocated 340 km upstream to a fragmented 35 km stretch of the river between two hydroelectric generating stations, where sturgeon were considered extirpated. This study assessed the translocation effort using telemetry (movement), demographics and genetic data. Within the first year, a portion of the radio-tagged translocated individuals dispersed out of the release area, and released radio-tagged individuals used different areas than individuals radio-tagged ten years later. Catches of juvenile lake sturgeon have increased over time, with 150 juveniles caught within the duration of this study. The reintroduced population had similar genetic diversity as the source population, with a marked reduction in effective population size (Ne). The results indicate that the reintroduction effort was successful, with evidence of successful spawning and the presence of juvenile lake sturgeon within the reintroduction site. Overall, the results suggest adult translocations may be a useful tool for re-establishing other extirpated lake sturgeon populations. Assessment of Potential Threats to Eastern Flowering Dogwood (Cornus florida) in Southern Ontario
In Canada, eastern flowering dogwood (Cornus florida L.) is an endangered tree that occurs only in the Carolinian forest of southern Ontario. Threats to this species include habitat fragmentation and the fungal pathogen dogwood anthracnose (Discula destructiva). I conducted a population genetic analysis using seven nuclear microsatellite markers to determine if fragmented populations are genetically isolated from one another and have low levels of genetic diversity. Genetic comparisons suggest on-going dispersal among sites and relatively high genetic diversity within most sites; however, smaller populations and younger trees were less genetically diverse. I also used linear mixed effects models to assess potential relationships between several ecological variables and the prevalence of dogwood anthracnose. Disease severity was higher in trees on shallow slopes and in larger trees; the latter also had higher likelihood of infection. Insights from this study will be important to incorporate into future management strategies. Assessing the population genetic structure of the endangered Cucumber tree (Magnolia acuminata) in southwestern Ontario using nuclear and chloroplast genetic markers.
Magnolia acuminata (Cucumber tree) is the only native Magnolia in Canada, where it is both federally and provincially listed as endangered.Magnolia acuminata in Canada can be found inhabiting pockets of Carolinian forest within Norfolk and Niagara regions of southwestern Ontario. Using a combination of nuclear and chloroplast markers, this study assessed the genetic diversity and differentiation of M. acuminata in Canada, compared to samples from the core distribution of this species across the United States. Analyses revealed evidence of barriers to dispersal and gene flow among Ontario populations, although genetic diversity remains high and is in fact comparable to levels of diversity estimated across the much broader range of M. acuminata in the USA. When examining temporal differences in genetic diversity, our study found that seedlings were far fewer than mature trees in Ontario, and in one site in particular, diversity was lower in seedlings than that of the adult trees. This study raises concern regarding the future viability of M. acuminata in Ontario, and conservation managers should factor in the need to maintain genetic diversity in young trees for the long-term sustainability of M. acuminata in Ontario. Assessing the environmental correlates of a lethal amphibian pathogen, Batrachochytrium dendrobatidis, in Ontario wetlands
Many emerging infectious diseases are caused by pathogens that possess free-living life stages, in which they interact with the environment directly rather than through the mediation of a host. These diseases represent major impediments to wildlife conservation; however, the dynamics of their interaction with the environment are poorly studied, often due to the difficulty of detecting these microscopic pathogens in environmental samples. One of these pathogens is Batrachochytrium dendrobatidis (Bd), a fungus that has been linked to declines in many amphibian species. In this thesis I use an emerging technique, environmental DNA detection (eDNA), to detect and quantify Bd in the water of southern Ontario (Canada) wetlands and examine its correlation with a variety of aspects of water quality, surrounding habitat, and seasonal timing. My purpose was to inform not only on potential environment-pathogen dynamics for Bd in northern environments, but to provide insight into the use of eDNA as a disease surveillance tool. I found that not only was there high geographic variation in Bd detection and intensity, but also high temporal variation within the same site on time scales as low as two weeks. While Bd prevalence was not strongly correlated with any of the variables tested, intensity showed strong correlation with canopy cover, with greater canopy cover over a waterbody correlating to lower Bd intensity. My results present several promising avenues for further examination of Bd in northern ecosystems, and indicate that, while caution is warranted in its implementation, eDNA may become an important tool in amphibian pathogen surveillance. Assessing the Potential of Permaculture as an Adaptation Strategy Towards Climate Change in Central Ontario
This thesis uses three approaches to assess the potential of permaculture in Central Ontario. This was done using a vegetable field trial and modelling programs to determine the effectiveness of permaculture to decrease negative impacts of climate change based on projected climate values derived from regional circulation models. The first approach showed no statistical difference (P<0.05) of applying varied volumes and combinations of organic amendments on crop yields. The second approach indicated permaculture may be a sustainable production system with respect to soil erosion when compared to traditional agricultural practices. The third approach was inconclusive due to the lack of quantitative literature on permaculture management impacts on biomass yields, soil carbon or nutrient retention, which were missing from basic and scientific literature searches. Assessing the Potential for Contamination of Lakes from Upwelling of Arsenic-Laden Groundwater Through Sediments
A bedrock fracture hosting arsenic (As) contaminated groundwater was suspected to be transported to Ramsey Lake, a drinking water resource for more than 50,000 residents of Sudbury, Ontario. A high resolution, spatial, water quality mapping technique using an underwater towed vehicle (UTV) was used to identify sources of upwelling groundwater into lake water and localize the upwelling As contaminated groundwater vent site. The top 7 cm of lake sediments (in-situ) at this vent site were observed to adsorb 93 % of the dissolved As, thus inhibiting lake water quality degradation from this contaminant source. Sediment samples from this location were used in laboratory experiments to assess the potential for this system to become a source of As contamination to Ramsey Lake water quality and elucidate As(III) fractionation, transformation and redistribution rates and processes during aging. Arsenic speciation is important because As(III) has been shown to be more toxic than As(V). To accomplish this a sequential extraction procedure (SEP) that maintains As(III) and As(V) speciation in (sub)oxic sediments and soils was validated for the operationally defined fractions: easily exchangeable, strongly sorbed, amorphous Fe oxide bound, crystalline Fe oxide bound, and the residual fraction for total As because the characteristics of the reagents required to extract the final fraction do not maintain As species. Batch reaction experiments using sediment spiked with As(III) or As(V) and aged for up to 32 d were sequentially extracted and analysed for As(III) and As(V). Consecutive reaction models illustrate As(III) is first adsorbed to the sediment then oxidized to As(V). Fractionation analyses show As(III) most rapidly adsorbs to the easily exchangeable fraction where it is oxidized and redistributes to the strongly sorbed and amorphous Fe oxide bound fractions. Oxidation of As(III) adsorbed to the amorphous and crystalline Fe oxide bound fractions is less efficient and possibly inhibited. Select samples amended with goethite provide evidence supporting Mn(II) oxidation is catalyzed by the goethite surface, thus increasing As(III) oxidation by Mn(III/IV) complexed with the strongly sorbed fraction. Although As immobilization through groundwater sediment interactions may be inhibited by increased ion activity, particularly phosphate or lake eutrophication, this threat in Ramsey Lake is likely low.
