Mutation of the B10 Tyrosine and E11 Leucine in Giardia intestinalis Flavohemoglobin
The flavohemoglobin in Giardia intestinalis (gFlHb) is the only known protozoan member of a protein class typically associated with detoxifying nitric oxide (by oxidation to nitrate) in bacteria and yeast. Mutants of the B10 tyrosine (Y30F) and E11 leucine (L58A), conserved residues thought to influence ligand binding, were expressed and studied using Resonance Raman (RR) spectroscopy. In the wild type protein, RR conducted using a carbon monoxide probe detects two distinct Fe-CO stretches associated with two different active site configurations. In the open configuration, CO does not interact with any polar side chains, while in the closed configuration, CO strongly interacts with one or more distal residues. Analysis of the Y30F mutant provided direct evidence of this tyrosine’s role in ligand stabilization, as it had only a single Fe-CO stretching mode. This stretching mode was higher in energy than the open conformer of the wild type, indicating a residual hydrogen bonding interaction, likely provided by the E7 glutamine (Q54). In contrast the L58A mutant had no effect on the configurational nature of the enzyme. This was unexpected, as the side chain of L58 sits atop the heme and is thought to regulate the access of distal residues to the heme-bound ligand. The similar spectroscopic properties of wild type and L58A suggest that any such regulation would involve rapid conformational dynamics within the heme pocket. Elemental Variation in Daphnia
Environmental variation can affect consumer trait expression and alter ecological and evolutionary dynamics in natural populations. However, although dietary nutrient content can vary by an order of magnitude in natural ecosystems, intra-specific differences in consumer responses to food quality have not been thoroughly investigated. Therefore, the purpose of my dissertation was to examine the influence of dietary nutrition and other environmental factors on consumer phenotypic variation using the freshwater cladoceran Daphnia. I conducted a series of complementary laboratory and field studies where I examined the effects of dietary phosphorus (P) content and additional biological/environmental variables (multi-elemental limitation, genetic variation, and temperature) on daphnid life-history, biochemistry, body elemental composition, and population growth. In general, phenotypic expression within a species varied significantly in response to all experimental variables, but the relative influence of each was highly context dependent. In my first chapter, I found that dietary P content and environmental calcium (Ca) concentrations both altered Daphnia body Ca:P ratios and growth rates of individuals and affected intrinsic rates of increase at the population level. However, food quality appeared to have a much larger effect on trait expression, and body Ca:P ratios were highly sensitive to other forms of dietary nutrient limitation. Next, I documented significant quantitative genetic variation and phenotypic plasticity in daphnid P content, growth, and P use efficiency of field collected animals grown across dietary P gradients. Trait expression was also influenced by genotype X diet interactions suggesting that consumer responses to dietary nutrient limitation can be heritable and may be adaptive in different nutrient environments. Finally, I found that temperature appeared to override food quality effects and decouple P metabolism in natural Daphnia populations, but total biomass production was affected by both dietary P content and temperature, depending on the nutrient content of the lake. Overall, my dissertation shows that consumer responses to nutrient limitation can vary significantly within a species and that changes in trait expression may be modified by other environmental variables. These results should be incorporated into existing stoichiometric models and used to investigate the eco-evolutionary consequences of consumer phenotypic variation in response to nutritional stress. Investigation of a Model for Stain Selection and a Robust Estimation for Area of Origin in Bloodstain Pattern Analysis
Bloodstain pattern analysis is fundamental to crime scene investigations by providing insight into the estimation of the area of origin of a blood source. The selection of bloodstains from an impact pattern is the foundation from which the estimation of the area of origin is derived. The purpose of this research was to develop a theoretical model for stain selection and establish a more robust estimation for the area of origin in the analysis of impact patterns. This model was developed by using quantitative data from marked zones on the impact pattern target surface. The minimum distance between the known blood source location and a single bloodstain line of trajectory was calculated for every bloodstain within each pattern investigated. It has been determined that the zone location of a bloodstain is relevant for improving accuracy, and that stains with all alpha angles can be used to calculate an area of origin estimation. Finally, research within this thesis also validated directional analysis and investigated the number of stains that may be required for a precise area of origin estimation. Politics of Muslim Intellectual Discourse in the West
The dissertation explores and defends the theory and practice of a Western-Islamic public sphere (which is secular but not secularist and which is Islamic but not Islamist), within which a critical Islamic intellectual universe can unfold, dealing hermeneutically with texts and politically with lived practices, and which, moreover, has to emerge from within the arc of two alternative, conflicting, yet equally dismissive suspicions defined by a view that critical Islam is the new imperial rhetoric of hegemonic orientalism and the opposite view that critical Islam is just fundamentalism camouflaged in liberal rhetoric. The Western-Islamic public sphere offers a third view, arising from ethical commitment to intellectual work, creativity, and imagination as a portal to the open horizons of history. Early Responses of Understory Vegetation to Above Canopy Nitrogen Additions in a Jack Pine Stand in Northern Alberta
Abstract Early Responses of Understory Vegetation After One Year of Above Canopy Nitrogen Additions in a Jack Pine Stand in Northern Alberta Nicole Melong Nitrogen (N) emissions are expected to increase in western Canada due to oil and gas extraction operations. An increase in N exposure could potentially impact the surrounding boreal forest, which has adapted and thrived under traditionally low N deposition. The majority of N addition studies on forest ecosystems apply N to the forest floor and often exclude the important interaction of the tree canopy. This research consisted of aerial NH4NO3 spray applications (5, 10, 15, 20, 25 kg N ha-1yr-1) by helicopter to a jack pine (Pinus banksiana Lamb.) stand in the Athabasca Oil Sands Region (AOSR) in northern Alberta, Canada. The main objective was to assess the impacts of elevated N after one year of treatment on the chemistry of understory vegetation, which included vascular plants, terricolous lichens, epiphytic lichens and a terricolous moss species. Changes in vegetation chemistry are expected to be early signs of stress and possible N saturation. Increased N availability is also thought to decrease plant secondary compound production because of a tradeoff that exists between growth and plant defense compounds when resources become available. Approximately 60% of applied N reached the ground vegetation in throughfall (TF) and stemflow (SF). Nitrate was the dominant form of N in TF in all treated plots and organic N (ON) was the dominant form of N in SF in all plots. The terricolous non-vascular species were the only understory vegetation that responded to the N treatments as N concentration increased with increased treatment. Foliar chemistry of the measured epiphytic lichens, vascular species, and jack pine was unaffected by the N treatments. Based on biomass measurements and N concentration increases, the non-vascular terricolous species appear to be assimilating the majority of TF N after one year. Vegetation from the high treatment plot (25 kg N ha-1yr-1) was compared to a jack pine forest receiving ambient high levels of N (21 kg N ha-1yr-1) due to its proximity to Syncrude mining activities. Nitrogen concentrations in plant tissues did not differ between the two sites; however, other elements and compounds differed significantly (Ca, Mg, Al, Fe). After one year of experimental N application, there were no environmental impacts consistent with the original N saturation hypothesis. Reshaping the Terms of Debate
The Reagan era instigated a fundamentally conservative shift in the political, economic and discursive climate of America. As Ronald Reagan is a highly divisive symbolic figure in American politics, much of the historiography of his presidency has been characterized by polarized interpretations. Over the past decade there has been a noticeable shift towards more favourable and triumphal interpretations of the Reagan era. This thesis seeks to analyze the ideological shifts that have characterized the trajectory of historical writings on the Reagan era. Through employing a careful textual analysis of key works by Michael Schaller, Gil Troy and Sean Wilentz, amongst others, this study demonstrates how historiography serves us less as an objective means of understanding the past and more so as an expanding collective historical artifact that illustrates the changing currents of intellectual and political discourse. Commonality of Enemies
Carlism and anarchism were revolutionary social movements that acquired significant popular support during the most intensive period of modernization in Spain (mid 19th to mid 20th centuries). It was noted but not well explored by contemporaries and historians that these enemies were similar in their hostility towards modernization and in their intense idealism. This thesis compares the two movements in order to determine the nature of their commonality and what this suggests about ideological enemies. A range of sources were consulted, including scholarship on modern Spain, biographical information on individuals who converted from Carlism to anarchism and contemporary print media. It was concluded that they were produced by the same destabilizing processes of disentailment and industrialization, which drew the working classes towards proposals that would have otherwise seemed implausibly utopian. The thesis further suggests that they were uniquely idealistic, in that they put moral integrity before the success of their cause. Assessing the Effects of Brief Interventions on Mood and Dream Imagery in Recovering Alcohol and Drug Addicts
Addiction can lead to a plethora of health, social and economical problems. Substances are used for mood regulation, and therefore, waking day mood is extremely important during recovery of alcohol and drug addiction. The current study examined the effects of Meditation and Sleep Mentation Therapy on mood levels. All participants were male, and currently in treatment programs or early stages of recovery from alcohol and drug addiction. Participants were tested for anxiety and depression and were asked to provide a recent dream, prior and post to participating in the intervention. Dreams were scored using Hall and Van de Castle guidelines for scoring imagery. Results are consistent with previous research in that mood levels changed over the course of the meditation period. Implications for future research are discussed as well as applications of Sleep Mentation Therapy and Meditation in clinical and applied practice. Habitat use and community structure of grassland birds in southern Ontario agro-ecosystems.
Most grassland bird populations are in decline, so it is becoming increasingly important to understand how they use agricultural field types and form their communities. I performed point counts in cultural meadow, intensive agriculture, and non-intensive agriculture areas in 2011 and 2012. Generalized linear models were used to determine the habitat relationships of six focal species. I found that non-intensive agriculture was used most often and intensive agriculture was often avoided, but there were exceptions which indicate habitat use can be species-specific. I determined in which habitats competition was likely occurring and which species pairs were competing in 2011. In 2012, I experimentally tested these relationships by introducing artificial competitors onto sites. By comparing presence-absence data from 2011 to 2012, I found evidence of habitat-mediated interspecific and conspecific attraction involving Bobolink and Grasshopper Sparrow. This research contributes to the current understanding of grassland bird community ecology and conservation. Aeolian Impact Ripples in Sand Beds of Varied Texture
A wind tunnel study was conducted to investigate aeolian impact ripples in sand beds of varied texture from coarsely skewed to bimodal. Experimental data is lacking for aeolian megaripples, particularly in considering the influence of wind speed on ripple morphometrics. Additionally, the modelling community requires experimental data for model validation and calibration. Eighteen combinations of wind speed and proportion of coarse mode particles by mass were analysed for both morphometrics and optical indices of spatial segregation. Wind tunnel conditions emulated those found at aeolian megaripple field sites, specifically a unimodal wind regime and particle transport mode segregation. Remote sensing style image classification was applied to investigate the spatial segregation of the two differently coloured size populations. Ripple morphometrics show strong dependency on wind speed. Conversely, morphometric indices are inversely correlated to the proportion of the distribution that was comprised of coarse mode particles. Spatial segregation is highly correlated to wind speed in a positive manner and negatively correlated to the proportion of the distribution that was comprised of coarse mode particles. Results reveal that the degree of spatial segregation within an impact ripple bedform can be higher than previously reported in the literature. Constraints on phenotypic plasticity in response to predation risk
Inducible defenses are plastic responses by an organism to the perception of predation risk. This dissertation focuses on three experiments designed to test the hypothesis that plastic ability is limited by energetic constraints. Chapter 1 provides a general introduction to phenotypic plasticity research and the theoretical costs and limitations affecting the expression of plastic traits. In Chapter 2, I tested the hypothesis that costs of early plasticity may be manifested by a reduced response to risk in later life stages. I found that amphibian embryos are able to detect and respond to larval predators, but that the energetic cost of those plastic responses are not equivalent among behavioural, growth, and morphological traits, and their expression differs between closely-related species. Chapter 3 explicitly examines the relationship between food resource availability and plasticity in response to perceived predation risk during larval development. Food-restricted tadpoles showed limited responses to predation risk; larvae at food saturation altered behaviour, development, and growth in response to predation risk. Responses to risk varied through time, suggesting ontogeny may affect the deployment of particular defensive traits. Chapter 4 examines the influence of maternal investment into propagule size on the magnitude of the plastic responses to predation risk in resulting offspring. I found that females in better body condition laid larger eggs and that these eggs, in turn, hatched into larvae that showed greater morphological plasticity in response to predation risk. Maternal investment can therefore affect the ability of offspring to mount morphological defenses to predation risk. Last, Chapter 5 provides a synthesis of my research findings, identifying specific factors constraining the plastic responses of prey to perceived predation risk. Overall, I found constraints on plastic responses imposed by the current environment experienced by the organism (resource availability), the prior experience of the organism (predator cues in the embryonic environment), and even the condition of the previous generation (maternal body condition and reproductive investment). Together, these findings both provide new knowledge and create novel research questions regarding constraints limiting phenotypic variation in natural populations. "Energetics" of Mycenaean Defense Works
This thesis examines the mobilization of labour required for fortification construction during the Late Helladic (LH) period of the Aegean Bronze Age. It adopts an "energetics" approach to architecture, as a framework for systematically calculating the labour costs of construction, and using such costs to infer relative differences in political power among groups and communities through the implied differences in labour control. Accordingly, construction costs were generated for thirty-six LH fortifications, located across seven distinct regional zones of the Greek mainland and Aegean Sea. These values were then compared and evaluated against what is known of the political geographies for each region, to measure the extent to which the mobilization of labour was a function of regional power in Late Bronze Age Greece. These assessments revealed that a wide range of variation existed among the sampled regions in terms of the strength and nature of this connection, underscoring the diversity in labour relations that developed throughout the Aegean during the LH period. The labour costs were also used to suggest specific systems of recruitment that may have been in place for mobilizing workers, and to argue that fortification construction would not have been particularly burdensome or demanding for certain local populations.
