Comparative phylogeography in conservation biology
Phylogeographic histories of taxa around the Great Lakes region in North America are relevant to a range of ongoing issues including conservation management and biological invasions. In this thesis I investigated the comparative phylogeographic histories of plant species with disjunct distributions and plant species with continuous distributions around the Great Lakes region; this is a very dynamic geographic area with relatively recent colonisation histories that have been influenced by a range of factors including postglacial landscape modifications, and more recently, human-mediated dispersion. I first characterized four species that have disjunct populations in the Great Lakes region: (Bartonia paniculata subsp. paniculata, Empetrum nigrum, Sporobolus heterolepis, and Carex richardsonii). Through comparisons of core and disjunct populations, I found that a range of historical processes have resulted in two broad scenarios: in the first scenario, genetically distinct disjunct and core populations diverged prior to the last glacial cycle, and in the second scenario more recent vicariant events have resulted in genetically similar core and disjunct populations. The former scenario has important implications for conservation management. I then characterized the Typha species complex (T. latifolia, T. angustifolia, T. x glauca), which collectively represent species with continuous distributions. Recent microevolutionary processes, including hybridization, introgression, and intercontinental dispersal, obscure the phylogeographic patterns and complicate the evolutionary history of Typha spp. around the Great Lakes region, and have resulted in the growing dominance of non-native lineages. A broader geographical comparison of Typha spp. lineages from around the world identified repeated cryptic dispersal and long-distant movement as important phylogeographic influences. This research has demonstrated that comparisons of regional and global evolutionary histories can provide insight into historical and contemporary processes useful for management decisions in conservation biology and invasive species. From Negation to Affirmation
Forensic scientific practice is conventionally understood as a solution to absence. With every technological advance the power and span of the archive grows and with it revives hopes of uncovering facts and locate bodies that might put genocide denial and/or negationism to rest. Destruction, however, continues to define the reality and conditions for testimony in the aftermath of mass atrocity. This means that even as forensic scientific practice grows in its capacity to presence that which was previously unpresentable, destruction and the concomitant destruction of archive require that we consider what it means to remember with and without the archive alike. This dissertation explores the impact of forensic science on cultural memory through a choice of two case studies (set in Kosovo and Srebrenica respectively) where forensic scientific methods were involved in the investigation of atrocities that were openly denied. This dissertation makes an agnostic argument that the biblical example of the empty tomb can serve as a paradigm to understand the terms of witnessing and testifying to absence in the era of forensic scientific investigations. Specifically, it posits the following theses with regards to the empty tomb: it is a structure and an event that emerges at the intersection of forensic science’s dual property as an indexical technique and as a witness function, it cannot be validated through historiographic or forensic scientific methods (it is un-decidable) and as such serves as a corrective the fantasy of the total archive, is represented in the contemporary genre of forensic landscape; and because it breaks with the forensic imperative, it compels alternative uses for testimony and memorial practices that need not be defined by melancholia as it can accommodate forms of testimony that are joyous and life affirming. Environmental Health Management Practices in Indigenous Communities
Environmental factors play a critical role in the health and well-being of people worldwide and the distribution of the burden of disease associated with environmental causes is disproportionately high in marginalized populations, including First Nations. In this way, environmental health problems are as much social and political problems as environmental and must be addressed as such. In Canada, the division of responsibilities for environmental health, in combination with the jurisdictional complexities of health and environment regulation and service provision on-reserve creates a First Nations environmental health management system with significant gaps. This research set out to explore the question: What are the current strengths and challenges in First Nations environmental health policy and management? A qualitative exploratory design organized in two stages and employing key informant interviews, document review and a community case study was used to examine this topic. In the first stage a review of existing programs and policies applicable to Ontario First Nations and a series of interviews with key experts on the topic in the province were conducted. A conceptual framework of the core elements affecting environmental health management in First Nations communities was developed and then applied to a case study with Mississauga First Nation in Northern Ontario. The framework included five core elements: Environmental Health Jurisdiction and Responsibility; Participation in Environmental Health Decision-Making; Access to Environmental Health Resources, Communication of Environmental Health Information; and, Role and Influence of Leadership. The findings indicate that "internal" issues, like community-based decision-making and support for environmental health initiatives seem to be least affected by the "external" issues such as access to federal funding. The "internal" issues were also shown to be critically important factors having impacts on environmental health management practices and policies in Mississauga First Nation. While there are countless barriers associated with the "external" factors that have significant impacts on environmental health management practices and policies, this research suggest that the "internal" factors can potentially be the most important factors in creating positive change in this area and as a result warrant further study in order to improve the state of environmental health issues in First Nations. Beyond Habitat
My objective was to understand how individual variation, in conjunction with variation in habitat, can affect individual and population-level variation in animal space use. I used coyotes (Canis latrans) as a model species to investigate the roles of hybridization, an inherited intrinsic factor, and spatial memory, a learned intrinsic factor, on space use. I used a diversity of methods and approaches, including meta-regression, multiple imputation, simulations, resource selection functions, step selection functions, net-squared displacement analysis, and survival analysis. A major contribution was my investigation of the performance of multiple imputation in a meta-regression framework in Chapter 2. My simulations indicated that multiple imputation performs well in estimating missing data within a meta-regression framework in most situations. In Chapter 3, I used published studies of coyote home range size in a meta-regression analysis with multiple imputation to examine the relative roles of hybridization and environmental variables on coyote home range size across North America. I found that hybridization with Canis species was a leading factor driving variation in coyote space use at a continental scale. In Chapter 4, I used telemetry data for 62 coyotes in Newfoundland, Canada, to investigate the influence of cognitive maps on resource use. I found that resident coyotes used spatial memory of the landscape to select or avoid resources at spatial scales beyond their immediate sensory perception relative to transient coyotes, presumably increasing their fitness. Taken together, my dissertation demonstrates that intrinsic factors, such as genetic ancestry and spatial memory, can have substantial influences on how animals use space at both individual and population levels, and at both a local and a continental scales. Characterization of frog virus 3 and its binding partner LITAF
Iridoviruses are large (120-200nm) double stranded DNA viruses that contain an icosahedral capsid. The iridoviridae family is composed of five genera that infect a wide range of poikilothermic vertebrates (Lymphocystivirus, Ranavirus and Megalocyivirus) and invertebrate hosts (Iridovirus, Chloriridovirus). Frog virus 3 (FV3) is a member of the Ranavirus genus, and is commonly used as a model system to study iridoviruses. I was interested in understanding virus-host interaction in FV3. I studied two viral genes, FV3 97R and FV3 75L. Here I demonstrate that 97R localizes to the endoplasmic reticulum (ER) at 24 hours post-transfection. However, at 35 hours post-transfection 97R localizes to the ER but also begins to form concentrated pockets, continuous with the nuclear membrane This study found that 97R possess a unique phenotype and that its localization to the ER is mediated through its C-terminus transmembrane domain. FV3 75L encodes an 84 amino acids protein. I showed that FV3 75L localizes to the early endosomes, while its cellular binding partner, LITAF, localizes to late endosome/lysosome. Interestingly, when FV3 75L and LITAF are co-transfected into cells, LITAF can alter the subcellular localization of FV3 75L to late endosome/lysosomes. A physical interaction between LITAF and FV3 75L was demonstrated through a pull-down assay and that a highly conserved domain found in both proteins may mediate the interaction. LITAF has been proposed to function in protein degradation, but there is still uncertainty on LITAF's specific role. I was interested in further characterizing LITAF and its implications in protein degradation and a neurodegenerative disorder. At least 9 mutations of LITAF are associated with Charcot-Marie-Tooth disease type 1C (CMT1C), which belongs to the group of most common heritable neuromuscular disorders, affecting approximately one in 2500 people. We show that LITAF mutants G112S and W116G mislocalize from the late endosome/lysosome to the mitochondria while the T49M and P135T mutants show partial mislocalization with a portion of the protein present in the late endosome/lysosome and a portion of the protein localized to the mitochondria. Since LITAF is believed to play a role in protein degradation, it is possible that the specific characteristics of CMT1C may occur though impaired degradation of Schwann cell membrane proteins, such as PMP22. I was able to show that when WT LITAF is present, there is a decrease in the PMP22 intracellular levels, which suggest that LITAF plays an important role in protein degradation, and also in other types of CMT. Insight into how mutations in LITAF cause CMT1C may not only help better understand cellular pathways, but also further elucidate the role LITAF's viral homolog FV3 75L during viral infection. Involvement of Endogenous Plant Hormones in The Regulatory Network of Fatty Acid Biosynthesis in Soybean Seed
The activities of phytohormones during the reproductive phase have been partially clarified in seed physiology while the biological role of plant hormones in oil accumulation during seed development has been investigated in part only. In this research, fatty acid (FA) contents and hormone profiles, including abscisic acid (ABA) and cytokinins (CKs) of seed samples in four different stages and comparing six soybean varieties have been investigated in order to examine the hypothesis that the endogenous plant hormones play important roles in FA production in soybean seeds. The FA contents increased significantly during this period while the hormone concentrations gradually declined towards the seed physical maturation. However, the interactions between FA contents and hormone profiles were complex and went beyond linear correlations. Hormone metabolism in the earlier stages of seed maturation period demonstrated numerous robust relationships with FA accumulations, as derived from several simple and multiple regression models in the determination of different FA contents. Evaluation of the effects of exogenous ABA and trans-Zeatin (tZ) on FA biosynthesis has revealed that ABA appears to be involved in the accumulations of unsaturated FAs while tZ participated in the synthesis of saturated and unsaturated FAs. Native Art as seen through Native Eyes
Since the end of the Second World War, artists of Native descent have engaged with the Fine Art world where their work has come to be placed in the category of Native art. As a result of my journey, I have come to realize that in the Fine Art world the term Native art tends to be associated with the practices of our ancestors in times past obscuring our contemporary nature. In the present day context, however, I see an evolution and will tell the stories of the artists I met, who became a part of my life and thus a part of my narrative to point out that the voices of contemporary artists of Native descent, when speaking of their work, demonstrate a modern form of Native creativity, pride and joy that needs to be properly recognized. While Native artists do respect our traditions and do deal with issues of importance to our communities, they also create their artwork using sophisticated and modern techniques. It is up to us to make our contemporary nature known far and wide. A storytelling approach based on the Michael Thrasher Medicine Wheel Teachings is employed to present the voices of our contemporary artists of Native descent who when speaking of their work create a rich and vibrant story of Native creativity, pride and joy. MSHKIKENH IKWE NIIN (I am Tutle Woman)
This dissertation offers the dibaajimowin (personal story) of my beading project, which I undertook to understand the enactment of Anishinaabe women’s knowledge in graduate research. Framed through the concept of a collective self, which is embedded within the Anishinaabe traditions of storytelling and beading, and drawing further from the aesthetics, processes, and teachings of these traditions so that I proceed in a good way, I am able to tell a story that is wholly Anishinaabe. Through the symbolic literacy present within the pieces of beadwork entitled:”Turtle Woman,” “Turtle Woman Meets Grandmother Moon,” “Turtle Woman Marries a Beaver,” and “Turtle Woman Slays the Big Fish,” which I present in the second half of this story, I explore the relationship among Anishinaabe women’s knowledges, self, identity, power relations, allyship, sovereignty and good governance in graduate research. I conclude that if graduate research is framed as an extension of an Anishinaabe space, an ethics of responsibility emerges, setting the stage for graduate research that is rooted in responsibility, contributing to efforts of Anishinaabe sovereignty and community wellness. Overall through my conclusion, as well as the process that I employ, I make contributions in the areas of Indigenous thought, Indigenous methodologies, Indigenous governance, feminism, critical theory, pedagogy, and ally theory. Conservation Genetics of Woodland Caribou in the Central Boreal Forest of Canada
Maintaining functional connectivity among wildlife populations is important to ensure genetic diversity and evolutionary potential of declining populations, particularly when managing species at risk. The Boreal Designatable Unit (DU) of woodland caribou (Rangifer tarandus caribou) in Ontario, Manitoba, and Saskatchewan has declined in southern portions of the range because of increased human activities and has been identified as 'threatened' by the Committee on the Status of Endangered Wildlife in Canada (COSEWIC). In this dissertation, I used ten microsatellite DNA markers primarily from winter-collected fecal samples to delineate genetic structure of boreal caribou in declining portions of the range and increase understanding of the potential influence of the non-threatened Eastern Migratory DU of woodland caribou on genetic differentiation. Eastern migratory caribou are characterized by large home ranges compared to boreal caribou and migrate seasonally into portions of the Boreal DU range. A regional- and local-scale analysis using the spatial Bayesian clustering algorithm in program TESS delineated four regional clusters and 11 local clusters, with the majority of local clusters occurring along the southern periphery of the range. One of those clusters in Ontario corresponded spatially with the seasonal overlap of boreal and eastern migratory caribou and was characterized by substantial admixture, suggesting that the two DUs could be interbreeding. Next, I decoupled the impacts of historical and contemporary processes on genetic structure and found that historical processes were an important factor contributing to genetic differentiation, which may be a result of historical patterns of isolation by distance or different ancestry. Moreover, I found evidence of introgression from a currently unsampled population in northern Ontario, presumably barren-ground caribou (R. t. groenlandicus). Finally, because our analysis suggested recent processes were also responsible for genetic structure, I used a landscape genetics analysis to identify factors affecting contemporary genetic structure. Water bodies, anthropogenic disturbance, and mobility differences between the two DUs were important factors describing caribou genetic differentiation. This study provides insights on where conservation and management of caribou herds should be prioritized in threatened portions of the boreal caribou range and may have implications for future delineation of evolutionarily significant units. Nunavik Inuit Knowledge of Beluga
Socio-ecological systems are inherently complex and marine mammals are fundamentally challenging to study. In the Arctic, marine mammals occupy a central ecological role, as nutrient cyclers and as a source of food and culture for Indigenous peoples. Inuit have developed a rich knowledge system, which has not been fully actualized in application in most Arctic research. Considering the need for the best available information in marine mammal ecology, the research question guiding this dissertation was: How can multiple methods and approaches be used to more effectively gather, understand, and represent Inuit Knowledge for an improved understanding of marine mammal ecology? The dissertation investigates this question using a case study of beluga in Nunavik (Arctic Quebec) drawing on the expertise of hunters and Elders to better understand complex questions in marine mammal ecology. The thesis uses a transdisciplinary approach to address the dissertation objective and is comprised of a general introduction, followed by four chapters formatted as journal manuscripts, and closes with an integrated discussion and conclusion. The first manuscript examines the contributions of Traditional Ecological Knowledge (TEK) of beluga to marine mammal literature. The second manuscript uses a sub-set of data gathered through participant mapping to apply a mapping method to explore how the spatial aspects of TEK could be better documented, analyzed, and represented. The third and fourth papers are based on the knowledge shared by hunters and Elders. The third explores the questions ‘why do beluga migrate?’ and ‘what factors influence beluga movement?’. The fourth investigates aspects of beluga foraging ecology. This dissertation makes methodological contributions through the application of kernel density estimators to participant maps as a method for transforming multiple mapped narratives into a quantitative database. The understandings shared by hunters and Elders make significant ecological contributions, particularly to foraging (e.g. diet composition and seasonal energy intake), and movement ecology (e.g. potential drivers of migration). Broadly these findings contribute to our collective understanding of beluga ecology and have implications for wildlife management. effects of Dissolved Organic Matter (DOM) sources on Pb2+, Zn2+ and Cd2+ binding
Metal binding to dissolved organic matter (DOM) determines metal speciation and strongly influences potential toxicity. The understanding of this process, however, is challenged by DOM source variation, which is not always considered by most existing metal speciation models. Source determines the molecular structure of DOM, including metal binding functional groups. This study has experimentally showed that the allochthonous-dominant DOM (i.e. more aromatic and humic) consistently has higher level of Pb binding than the autochthonous-dominant DOM (i.e. more aliphatic and proteinaceous) by more than two orders of magnitude. This source-discrimination, however, is less noticeable for Zn and Cd, although variation still exceeds a factor of four for both metals. The results indicate that metal binding is source-dependent, but the dependency is metal-specific. Accordingly, metal speciation models, such as the Windermere Humic Aqueous Model (WHAM), needs to consider DOM source variations. The WHAM input of active fraction of DOM participating in metal binding (f) is sensitive to DOM source. The commonly-used f = 0.65 substantially overestimated the Pb and Zn binding to autochthonous-dominant DOM, indicating f needs to be adjusted specifically. The optimal f value (fopt) linearly correlates with optical indexes, showing a potential to estimate fopt using simple absorbance and/or fluorescence measurements. Other DOM properties not optically-characterized may be also important to determine fopt, such as thiol, which shows strong affinity to most toxic metals and whose concentrations are appreciably high in natural waters (< 0.1 to 400 nmol L-1). Other analytical techniques rather than Cathodic Stripping Voltammetry (CSV) are required to accurately quantify thiol concentration for DOM with concentration > 1 mg L-1. To better explain the DOM-source effects, the conditional affinity spectrum (CAS) was calculated using a Fully Optimized ContinUous Spectrum (FOCUS) method. This method not only provides satisfactory goodness-of-fit, but also unique CAS solution. The allochthonous-dominant DOM consistently shows higher Pb affinity than autochthonous-dominant DOM. This source-discrimination is not clearly observed for Zn and Cd. Neither the variability of affinity nor capacity can be fully explained by the variability of individual DOM properties, indicating multiple properties may involve simultaneously. Together, the results help improve WHAM prediction of metal speciation, and consequently, benefit geochemical modelling of metal speciation, such as Biotic Ligand Model for predicting metal toxicity. Investigation of fugitive dust emissions from nepheline syenite mine tailings near Nephton, Ontario
A set of experiments was designed to investigate the factors—atmospheric and surficial—controlling fugitive dust emissions from the tailings ponds of UNIMIN Canada, a mining company that extracts and produces nepheline syenite (feldspar) at two adjacent sites (Nephton and Blue Mountain) located north of Havelock, Ontario. Using wind tunnel measurements, the combined influence of relative humidity and temperature (represented by the absolute matric potential, |Ψ|) on dust emission was quantified and modelled. About 300 experimental runs were conducted under various conditions of wind speed (4.5-6.25 ms-1), temperature (0-30oC) and relative humidity (10-70%). Generally, dust flux decreased as a logarithmic function of matric potential, with dust emission strongly suppressed for RH > 60% or |Ψ|<70 MPa. Field measurements also confirmed the role of relative humidity in suppressing dust emission. Irrigation, which is widely used by mines to control dust emissions, reduced ambient dust concentration at the study site only about 60% of the time, with the highest mitigation efficiencies (average of 90%) occurring when the total depth of water applied intermittently over a few hours was greater than 10 mm. In the absence of emergent vegetation, the terrestrial laser scanning (TLS) technique proved to be a promising method for detecting and estimating both spatial and temporal moisture content changes in the field environment, particularly for the very thin surface layer, which is the most important layer for dust emission. It is hoped that the results from this study will help mines to optimize their dust management programs for the range of climate and topographic conditions found at their sites, and also serve as a source of useful information and input data for atmospheric dispersion models, such as AERMOD and CALPUFF, whose accuracy depends on the quality of the input data such as the emission rate.
