Effect of Nitrosative Stress on Heme Protein Expression and Localization in Giardia Intestinalis
The parasitic protist Giardia intestinalis has five heme proteins: a flavohemoglobin and several isotypes of cytochrome b5. While the flavohemoglobin has a role in counteracting nitric oxide, the functions of the cytochromes (gCYTb5s) are unknown. In this study, the protein level and cellular localization of three gCYTB5 isotypes (gCYTb5-I, II and III) and flavohemoglobin were examined in Giardia trophozoites exposed to three nitrosative stressors at two different concentrations: nitrite (20 mM, 0.5 mM); GSNO (2 mM, 0.25 mM) and DETA-NONOate (2 mM, 0.05 mM). An increase in protein levels was observed for gCYTb5-II with all stressors at both concentrations. However, the effects of these nitrosative stressors on gCYTb5-I and III were inconclusive due to the variation among the replicates and the poor detection of gCYTb5- III on western blots. The protein level of the flavohemoglobin also increased in response to the three stressors at the low concentrations of stressors that were tested. Only the cellular localization of gCYTb5-I changed in response to nitrosative stress, where it moved from the nucleolus to the nucleus and cytoplasm. This response was extremely sensitive and occurred at the lower doses of the three stressors, suggesting that gCYTb5-I may be involved in a nucleolar- based stress response. Author Keywords:
Contemporary adaptive shifts in the physiology and life history of Pumpkinseed (Lepomis gibbosus) introduced into a warm climate
Contemporary evolution has the potential to help limit the biological impact of rapidly changing climates, however it remains unclear whether wild populations can respond quickly enough for such adaptations to be effective. In this thesis, I used the introduction of native North American Pumpkinseed (Lepomis gibbosus) into the milder climate of Europe over 140 years ago, as a 'natural' experiment to test for contemporary evolution to a change in climate in wild populations. In 2008, four outdoor pond colonies were established in central Ontario using adult Pumpkinseed from two native Canadian populations, and two non-native populations from northeastern Spain. By raising native and non-native Pumpkinseed within a common environment, this design minimized the impact of phenotypic plasticity on differential trait expression, and allowed me to interpret differences in the phenotype among pond-reared Pumpkinseed as evidence of genetic differences among populations. I demonstrated that Canadian and Spanish Pumpkinseed have similar thermal physiology except when acclimated to seasonally warm temperatures; trait differences are consistent with Spanish Pumpkinseed being better adapted to a warmer climate. Populations also had similar overwintering ecology, however some differences, such as higher survival under starvation conditions and greater energetic benefits associated with winter feeding, indicated that Canadian populations are better adapted to harsh winter conditions typical of the native range. Finally, I determined that the relatively fast life history expressed in wild European Pumpkinseed is largely driven by plastic responses to the local environment; however, the higher reproductive investment by European populations has a genetic basis. Most climate change research considers taxa that are expected to be negatively impacted by warming: my research demonstrates that even warm-tolerant taxa that are unlikely to experience strong climatic selective forces can respond to a warming environment through evolutionary changes. The potential for adaptive contemporary evolution in warm-tolerant taxa should be taken into account when predicting future ecosystem effects of climate change, and when planning management strategies for species introduced into novel climates. Lost Landscapes of the Kawarthas
The Kawartha Lakes region of south-central Ontario is dominated by water bodies and rivers, where humans are known to have lived since at least 10,500 years ago, only shortly after the retreat of glaciers from the region. Since this time, water levels within the region have changed dramatically as a result of various geophysical, climatological, and human-induced-phenomenon, leaving modern water levels at a maximum high-stand. While it is acknowledged within the local archaeological community that these hydrological dynamics have resulted in the inundation of much of the region’s past terrestrial and culturally active landscapes, cultural research into the region’s lakes and waterbodies have to date been very few and limited in scale. The subject of this thesis concerns a cultural assessment of the inundated landscapes around an island within Pigeon Lake of the Kawartha Lakes region, known as Jacob Island. Using a series of integrated methods including bathymetric modeling, shoreline and ecological reconstruction, and in-water-visual artefact survey, the goals of this research relate to illuminating the nature of the Kawartha Lakes region’s underwater archaeological record and associating specific cultural occupations and land-use strategies with Jacob island’s inundated landscapes. Habitat selection by sympatric Canada lynx (Lynx canadensis) and bobcat (Lynx rufus)
Range expansion by the bobcat (Lynx rufus) may be contributing to range contraction by the Canada lynx (Lynx canadensis), but interactions between them are not well understood. To investigate the potential for competition, I conducted a literature review of hierarchical habitat selection by these two species. I determined that the lynx and the bobcat select different resources at the first and second orders, and that the fourth order is under-studied compared to higher orders. I therefore conducted a snow-tracking study of fine-scale habitat selection by lynx and bobcat in an area of sympatry in northern Ontario. I found that the two species selected similar resources at the fourth order, but appeared to be allopatric at the level of the home range. These results suggest that competition is unlikely to occur between lynx and bobcat, and other factors should be considered as more probable causes of the lynx range contraction. Temporo-spatial patterns of occupation and density by an invasive fish in streams
Since its introduction to North America in the 1990s, the Round Goby has spread throughout the Great Lakes, inland through rivers and is now moving into small tributary streams, a new environment for this species in both its native and invaded ranges. I explored density and temporal occupation of Round Gobies in four small streams in two systems in south-central Ontario, Canada in order to determine what habitat variables are the best predictors of goby density. Two streams are tributaries of Lake Ontario and two are tributaries of the Otonabee River, and all of these streams have barriers preventing upstream migration. I found that occupation and density differed between the systems. In the Otonabee River system, Round Gobies occupy the streams year round and the most important factor determining adult density is distance from a barrier to upstream movement, with the entire stream occupied but density highest next to the barriers. In the Lake Ontario system, density is highest at mid-stream and Round Gobies appear to occupy these streams mainly from spring to fall. Adult density in Lake Ontario tributaries is highest in sites with a high percentage of cobble/boulder and low percentage of gravel substrate, while substrate is less important in Otonabee River tributaries. Occupation and density patterns may differ due to contrasting environmental conditions in the source environments and distance to the first barrier preventing upstream movement. This study shows diversity in invasion strategies, and provides insight into the occurrence and movement patterns of this species in small, tributary streams. Molecular Composition of Dissolved Organic Matter Controls Metal Speciation and Microbial Uptake
Aquatic contaminant mobility and biological availability is strongly governed by the complexation of organic and inorganic ligands. Dissolved organic matter (DOM) is a complex, heterogeneous mixture of organic acids, amino acids, lipids, carbohydrates and polyphenols that vary in composition and can complex to dissolved metals thereby altering their fate in aquatic systems. The research conducted in this doctoral dissertation addresses 1) how DOM composition differs between phytoplankton taxa and 2) how DOM composition affects metal speciation and its subsequent microbial bioavailability in laboratory and field conditions. To accomplish this, a series of analytical methods were developed and applied to quantify thiols, sulphur containing DOM moieties, and the molecular composition of DOM. The works presented in this thesis represents one of the first comprehensive and multipronged analyses of the impact of phytoplankton metabolite exudates on microbial metal bioavailability. This dissertation demonstrated the analytical versatility of high-resolution mass spectrometry as a tool for compound specific information, as well as having the capabilities to obtain speciation information of organometallic complexes. The work presented in this PhD strengthens the understanding compositional differences of both autochthonous and allochthonous DOM and their effects on metal biogeochemistry. Socioloegal Mediation of Rave Sound System Technologies
The central scholarly contribution of this dissertation develops through bringing the theories of Michel Foucault to bear in a sociolegal study of rave culture's criminalization by the United Kingdom's 1994 Criminal Justice and Public Order Act. My methodology develops rave as a cultural keyword. This keyword navigates through a quasi-materialist definition of rave as a cultural codification of sound system technologies. I theorize the way in which sociocultural discourse indexes aestheticized representations and the cultural mythologies that rave sound system's technical mediation generate. These ideas trace the facticity of the legal documentation of rave’s criminalization. I inform this sociolegal history by situating Foucault's work on the genealogy of liberalism as a practical toolkit for associating the legal discourse on rave culture with the genealogy of festival. This opens up a dialogue with the work of Mikhail Bakhtin's theorizing of the festival’s ambivalent political climate. Such ideas are useful in documenting rave as an enduring mimicry of the tension between State and civil society. Pieter Bruegel the Elder’s 1559 painting, “The Fight Between Carnival and Lent”, captures this tension beautifully. The aptness of reading rave's criminalization in relation to Bruegel’s portrayal of landscape is accomplished by returning to Foucault, who defines liberalism's political technologies in relation to Judaeo-Christian precedents. I explore how these political technologies, pastoral power in particular, are helpful in tracing rave's genealogical relation to the festival's sociotechnical cartography. How Abiotic and Biotic Factors Can Alter the Competitive Landscape in an Aggressive Species Complex (Genus
Competition is known to impact population dynamics through both indirect and direct interactions, and direct interactions can often lead to injury in one or both parties. As such, response to injury through tissue regeneration can be important for surviving post-competitive interaction. However, the impacts of outside factors like temperature and genome size (e.g. polyploidy) are not well studied, especially in syntopic systems. We addressed this knowledge gap by comparing regeneration rates of diploid Ambystoma laterale and triploid unisexual Ambystoma at two ecologically-relevant temperatures. Environmental factors appeared to have stronger effects on regeneration than ploidy level, but overall mass was impacted more strongly by ploidy level. Interestingly, there was an interaction between temperature and time within unisexuals that was absent when comparing different ploidy levels, implying temperature has a more complex effect on polyploids. This study supports the hypothesis that polyploid organisms are better equipped to respond to shifts in their environments, which can give them a competitive advantage at the northern range limit of this species complex. What’s the trouble with women? Fostering female engagement in substance abuse programming
Although Canada’s healthcare system is designed for everyone to access services regardless of the person’s gender, age, or income, there are significant barriers for individuals accessing substance abuse services that live in areas outside of urban centres (Adbool, et al., 2017; Hardill, 2011). Women are particularly stigmatized by the lack of anonymity in smaller communities and often avoid engaging in substance abuse programs (Ashley, Marsden, & Thomas, 2003). The aim of the current thesis was to explore RedPath, a grassroots initiative in Port Hope, Ontario, geared to engaging individuals and encourage them to participate in substance abuse programming. This initiative employs a member from the community, called an Activator, who is tasked with engaging their peers. A qualitative study was conducted to explore the role of a hired RedPath Activator in facilitating access of female community members with substance abuse issues to services in the Port Hope community. Her role in supporting women was a specific interest, as the selection of a female Activator was a strategy to support the engagement of women to the program. The data was analyzed using a thematic content analysis approach. The most significant of these themes were (1) barriers and challenges in the community and (2) building trust to facilitate engagement and maintain attendance in the program. Do birds of a feather flock together
Populations have long been delineated by physical barriers that appear to limit reproduction, yet increasingly genetic analysis reveal these delineations to be inaccurate. The eastern and mid-continent populations of sandhill cranes are expanding ranges which is leading to convergence and warrants investigation of the genetic structure between the two populations. Obtaining blood or tissue samples for population genetics analysis can be costly, logistically challenging, and may require permits as well as potential risk to the study species. Non-invasively collected genetic samples overcome these challenges, but present challenges in terms of obtaining high quality DNA for analysis. Therefore, methods that optimize the quality of non-invasive samples are necessary. In the following thesis, I examined factors affecting DNA quality and quantity obtained from shed feathers and examined population differentiation between eastern and mid-continent sandhill cranes. I found shed feathers are robust to environmental factors, but feather size should be prioritized to increase DNA quantity and quality. Further, I found little differentiation between eastern and mid-continent populations with evidence of high migration and isolation-by-distance. Thus, the two populations are not genetically discrete. I recommend future population models incorporate migration between populations to enhance our ability to successfully manage and reach conservation objectives. Hoop Dance Project
This dissertation explores a 2017 elementary school Hoop Dance project that was organized by a white music teacher, and taught by an Indigenous artist in Peterborough, Ontario. It aims to respond to the Truth and Reconciliation Commission Calls to Action, numbers 10 and 63, which ask the federal government to sufficiently fund legislation that incorporates the following principles: “… developing culturally appropriate curricula” (p. 2), and “building student capacity for intercultural understanding, empathy, and mutual respect” (p. 7). The dissertation asks the question: In what ways will a seven-week Indigenous Hoop Dance Unit, taught by an Indigenous performing artist and facilitated by a white school teacher, contribute to reconciliation in an elementary school classroom in Ontario? I am the teacher in this study and have worked at this elementary school for five years. Throughout the project, I acted as facilitator, participant, and researcher, while Indigenous dancer and instructor Beany John planned and delivered the Hoop Dance content. Theoretically, the dissertation is organized around the Anishinabek seven grandmother/grandfather teachings, as taught by Ojibwe/Odawa educator and author Pamela Toulouse (2011). I believe that these seven traditional teachings are a meaningful basis upon which to build the project, not only because they inform Indigenous knowledge in the arts, but also because frequent reflection and referral to the teachings help remind me to remain connected to the “higher” purpose of the research throughout the project, which is to further the reconciliation process in Canada, and more broadly, to benefit society. Regarding methodology, I use arts based research (Leavy, 2015) and a constructivist grounded theory analysis, which embraces the subjectivity and positionality of the researcher (Creswell, 2012). The overall conclusion of the dissertation is that although the Hoop Dance project did not significantly address issues of Indigenous sovereignty in education nor our shared inherited legacy of colonial harm, it was a constructive step in the reconciliation project, largely due to the contributions of Beany John, whose teaching gently unsettled conventional educational practice at our school. Assessing Quality of Life for the Urban Inhabitants of Classical Angkor, Cambodia (c. 802-1432 CE)
This thesis examines the interrelationship of urban planning and population health at the site of Angkor (c. 802-1432 CE), the capital city of the Classical Khmer state, now found within modern-day Cambodia. The inhabitants of Angkor developed a settlement strategy that relied on the dispersal of water management features, rice fields, temples and residential areas, to best utilize the spread-out environmental resources of the surrounding monsoon-forest climate. Thus, the main question to be answered by this thesis is this: did the city-planning practice of dispersed, low-density agrarian urbanism promote resilience against the disease hazards associated with tropical environments? To answer this question, methods involved creating assessing environmental and socio-cultural factors which habituated the urban inhabitants of Angkor’s relationship to disease hazards. The results of this assessment demonstrate that it was not until the last stages of Angkor’s urban development, when non-farming members of the population were concentrated into the “core” area of temples within city, that the city’s inhabitants’ vulnerability to infectious disease increased. As the city took a more compact settlement form, it was not as environmentally compatible as the earlier dispersed pattern. Significantly, archaeological case studies such as this can illustrate the long-term development and end-result of urban planning to deal with disease hazards, both in terms of everyday occurrences, as well as during crisis events, which has important implications for contemporary research on environmental disasters today.
