Agony of Writing Or Ambivalent Reversal In Baudrillard's Stylistic Metamorphoses
Following Baudrillard's conceptual and stylistic shift of the mid-70s, this thesis argues that said shift is accounted for by understanding the ontological quandary Baudrillard found himself in after developing a theoretical agonism impossible to divorce from the practice of writing. By tracing the conceptual metamorphoses of key terms including semiotic ambivalence, symbolic exchange and theoretical writing itself as a total agonistic process, this thesis demonstrates that theory is not reducible to epistemic production but is rather the contentious site of challenge and aesthetic (dis)appearance. Each chapter examines a conceptual tension revealing insoluble, conflicting social forms. These forms reveal the reversibility Baudrillard finds at work in all social phenomena. These culminate in a chapter that tackles Baudrillard's writing itself as a social form that endeavours to embody the agonistic theoretical concept as a process rather than remaining a representation, or commentary on, ambivalent social conflict. Youth Justice in Canada
Strategies to reduce youth crime have been extensively researched and custody is not found to be effective. In the past, custody was a frequently used sentence, and while under the YOA the number of youth in custody was four times higher than that of adults in Canada. The use of custody sentences in Ontario has decreased in recent years, however; it remains above the Canadian average. Currently, alternatives to custody are also being implemented. This study aimed to gather lived experiences of those with firsthand experience in the youth justice system (offenders and staff). These individuals have working knowledge of effective practices for reducing recidivism. Eighteen semi-structured interviews were conducted. Interviews were coded and analysed using Interpretive Phenomenological Analysis. A number of themes emerged, including various views on the benefits of custody, the importance of relationships, challenges of the job and the need for increased focus on prevention. Exonic Trinucleotide Microsatellites
Trinucleotide repeats (TNRs) are a class of highly polymorphic microsatellites which occur in neutral and non-neutral loci and may provide utility for individual- and population-identification. Exonic trinucleotide motifs, in particular, offer additional advantages for non-human species that typically utilize dinucleotide microsatellite loci. Specifically, the reduction of technical artifacts, greater separation of alleles and greater specificity of amplification products leading to more efficient multiplexing and cross-taxa utilization. This study aims to identify and characterize polymorphic trinucleotide repeats and conserved primer sequences which are conserved across Cervidae (deer) species and their potential for individual identification in forensic wildlife investigations. Chapter one provides a broad introduction to trinucleotide microsatellites, chapter two deals with data-mining TNRs and chapter three applies the identified TNRs as genetic markers for individual identification. Results demonstrate proof-of-concept that exonic TNRs are capable of giving random match probabilities low enough to be employed in individual identification of evidentiary samples. Nature without Balance
This thesis critically analyses the connection between ideology and nature, and in particular, aims to reflect on the dominant discourses on the topic of ecological crisis. The ecological thought framework that I adhere to rests on a combination of Frankfurt School and Žižekian theories. This combination is not without serious tensions and deviations; however, central to this project are the ways in which their respective works extensively critique ideology, and propose subversive alternatives to and new meanings of how we can conceptualize nature without domination. Dominant ideas and critiques of nature and natural history emerged during the Enlightenment era, and as Adorno argues, fell victim to a “reduction ad hominem,” or the claim that in order to free oneself, one must dominate, appropriate, and master nature. I claim that the extreme choices in environmental politics today - namely organic populism on one hand and increased technological intervention on the other - fail to account for the ways ‘nature’ is a socio-historical construct, and moreover, is situated within a false reality wherein the ‘essence of existence’ is reduced to technological mastery. What we encounter in this cautionary armoury of paradoxical approaches to nature, then, is the ideological currents of established belief systems. By exposing the illusions within the concept nature, such as the argumentative persuasion that there exists an inherent balance, the elementary cell of ideology reveals itself alongside revolutionary possibilities. Characterization of frog virus 3 and its binding partner LITAF
Iridoviruses are large (120-200nm) double stranded DNA viruses that contain an icosahedral capsid. The iridoviridae family is composed of five genera that infect a wide range of poikilothermic vertebrates (Lymphocystivirus, Ranavirus and Megalocyivirus) and invertebrate hosts (Iridovirus, Chloriridovirus). Frog virus 3 (FV3) is a member of the Ranavirus genus, and is commonly used as a model system to study iridoviruses. I was interested in understanding virus-host interaction in FV3. I studied two viral genes, FV3 97R and FV3 75L. Here I demonstrate that 97R localizes to the endoplasmic reticulum (ER) at 24 hours post-transfection. However, at 35 hours post-transfection 97R localizes to the ER but also begins to form concentrated pockets, continuous with the nuclear membrane This study found that 97R possess a unique phenotype and that its localization to the ER is mediated through its C-terminus transmembrane domain. FV3 75L encodes an 84 amino acids protein. I showed that FV3 75L localizes to the early endosomes, while its cellular binding partner, LITAF, localizes to late endosome/lysosome. Interestingly, when FV3 75L and LITAF are co-transfected into cells, LITAF can alter the subcellular localization of FV3 75L to late endosome/lysosomes. A physical interaction between LITAF and FV3 75L was demonstrated through a pull-down assay and that a highly conserved domain found in both proteins may mediate the interaction. LITAF has been proposed to function in protein degradation, but there is still uncertainty on LITAF's specific role. I was interested in further characterizing LITAF and its implications in protein degradation and a neurodegenerative disorder. At least 9 mutations of LITAF are associated with Charcot-Marie-Tooth disease type 1C (CMT1C), which belongs to the group of most common heritable neuromuscular disorders, affecting approximately one in 2500 people. We show that LITAF mutants G112S and W116G mislocalize from the late endosome/lysosome to the mitochondria while the T49M and P135T mutants show partial mislocalization with a portion of the protein present in the late endosome/lysosome and a portion of the protein localized to the mitochondria. Since LITAF is believed to play a role in protein degradation, it is possible that the specific characteristics of CMT1C may occur though impaired degradation of Schwann cell membrane proteins, such as PMP22. I was able to show that when WT LITAF is present, there is a decrease in the PMP22 intracellular levels, which suggest that LITAF plays an important role in protein degradation, and also in other types of CMT. Insight into how mutations in LITAF cause CMT1C may not only help better understand cellular pathways, but also further elucidate the role LITAF's viral homolog FV3 75L during viral infection. Understanding Dimensions of Environmental Sustainability in a Northern Indigenous Context
Although the concept of environmental sustainability has become increasingly popular, the literature offers little practical guidance to direct priorities or actions to support environmental sustainability in northern Indigenous communities. A case study in Hopedale, Nunatsiavut, and a systematic literature review was undertaken to understand: 1) what aspects of the local environment are of value to a northern Indigenous community; and 2) what does existing literature identify as key elements of a community-based approach to monitor valued aspects of the environment in a northern Indigenous context. Hopedale residents spoke to the importance of going off on the land and identified a number of categories of places in their local environment of importance to them, including: 1) valued areas for human-use, 2) areas to protect, 3) areas of environmental concern, and 4) areas to monitor. The systematic literature review highlighted trends on community-based monitoring (CBM) publications, and identified key 13 elements of CBM approaches that are pertinent to northern Indigenous communities. Insights from this study will inform environmental planning and management in the case community of Hopedale, as well as offer guidance to enhance current and future CBM activities in the North and elsewhere. Sexual Dimporphism and Population Dynamics of Sub-Arctic Breding Dunlin (Calidris alpina hudsonia) near Churchill, Manitoba, Canada
Around the world, many populations of migratory shorebirds appear to be declining. Conservation strategies to reverse declining trends rely on, among other information, a firm understanding of breeding ground population dynamics. From 2010 to 2014, I studied a breeding population of Dunlin (Calidris alpina hudsonia) near Churchill, Manitoba using mark-recapture methods. I found that females were significantly larger than males. I subsequently developed an equation based on morphological features that successfully classified 87.1% of females and 92.6% of males, sexed using molecular techniques. Using program MARK, I quantified the annual apparent survival of adults (± SE) within the breeding population (0.82 ± 0.063 for males, 0.73 ± 0.12 for females). Transient adults made up a significant percentage of the female population (32%, P = 0.011), but non-significant in the male population (12%, P > 0.05). Re-sight rate was high for both sexes, and ranged from 0.86-0.90 per year. Sex, year, and nest initiation date were the factors that had the greatest influence on annual returns. There was no significant difference in mean inter-annual nest site distance between sexes (male: 82.01 ± 13.42 m, female: 208.38 ± 55.88 m, P > 0.05). The high survival estimates obtained in this study suggest that the breeding population is stable and may not be contributing to suspected population declines within the subspecies. Assessment of Corporate Social Responsibility Compliance
The modern world faces a number of social, economic, and environmental sustainability challenges. Since businesses are assumed to have a role in causing such problems, they must also play a role in finding solutions. In Canada, the extent to which corporate social responsibility is institutionalized in the oil and gas industry remains a contentious issue among stakeholders. This study examines the extent of corporate social responsibility compliance in oil and gas corporations through an assessment of the corporate social responsibility reporting of two oil and gas organizations. Comparative analysis was used to determine each firm’s operational level compliance with Global Reporting Initiative (GRI) guidelines in terms of corporate social responsibility reporting. The study showed that firms' levels of compliance with social, economic, and environmental responsibility are unequal. As a result, a five-part mechanism is recommended to strengthen corporate social responsibility in the industry. It’s a Mad, Mad, Mad, Mad World
The purpose of this study was to understand the lived experiences of mental health and illness in Kuwait. Twenty-six participants were interviewed, including mental health professionals, family physicians, and service users. Findings suggest that inequality, oppression, and human rights violations may drive mental health issues in Kuwait. However, rather than addressing these factors, many healthcare providers are endorsing psychological testing and psychiatric medication, which may be resulting in the same iatrogenic (physician-induced) drug dependence that is seen in North America. An analysis of mainstream psychological theory, research and practice is provided, along with a bioethical critique of the World Health Organization’s efforts to reduce the global ‘burden’ of mental disorders. This study cuts across disciplinary boundaries and 1) supports medical anthropologists’ criticisms of the ‘advancement’ of global mental healthcare; 2) provides participant-driven, community-based alternatives that are specific to Kuwait; and 3) informs culturally defined notions of ‘care’ and ‘ethics’. Mitogenome characterization of the shortnose sturgeon (Acipenser brevirostrum) for international trade validation of aquaculture-reared caviar
Identifying the population origin of aquaculture-reared caviar is crucial for both conservation and management strategies of farmed fish but could also facilitate international trade of a CITES regulated product. Shortnose sturgeon (Acipenser brevirostrum) is the main source of caviar production in Atlantic Canada, from Breviro Caviar Inc. aquaculture facility. Shortnose sturgeon are also listed as a species-at-risk under the Species At Risk Act. Currently there is no genetic method for delineating wild from aquaculture-reared caviar. By targeting the mitochondrial genome (mitogenome) using novel long-range PCR primers and next-generation sequencing (NGS) methods we have successfully sequenced the full mitogenome of 37 shortnose sturgeon. The purpose of this study was to increase the resolution of diagnostic variation among populations and to validate Canadian aquaculture-reared stock from wild US populations. Results provided a previously unobserved novel control region haplotype in high frequency within both the aquaculture-reared and Saint John River wild sample sets. Similar frequencies were observed with whole mitogenome haplotypes. Diagnostic mitochondrial lineage found in high frequency within the captive Breviro Caviar Inc. population has the potential to allow caviar product from Breviro Caviar Inc. to be distinguished from protected US shortnose sturgeon populations. The application of full mitogenomic characterization provides the potential to further resolve differences between aquaculture and natural Canadian shortnose sturgeon stocks, US/Canadian populations and to contribute to future conservation strategies. Future research identifying signatures of selection on the mitogenome between captive and wild populations and across latitudinal gradients found within the species range. These novel methods have produced a proof-of-concept to provide a "farm-to-fork" validation and ecobrand of Breviro Caviar Inc. product and its aquaculture origin to support importation into US caviar markets. Passive sampling of indoor and outdoor atmospheric nitrogen dioxide in the greater Toronto area
The reliability and performance of four passive sampler membrane coatings specific to nitrogen dioxide (NO2) were evaluated through co-exposure at multiple Ontario Ministry of Environment and Climate Change (OMOECC) active monitoring stations. All four coatings performed relatively similar under a wide range of meteorological conditions, notably showing exposure-specific atmospheric uptake rates. Further, indoor and outdoor atmospheric concentrations of NO2 (a marker of traffic-related air pollution) were evaluated at multiple elementary schools in a high-density traffic region of Toronto, Ontario, using a Triethanolamine based passive sampler membrane coating. Samplers were also co-exposed at OMOECC active monitoring stations to facilitate calibration of exposure-specific atmospheric uptake rates. Indoor NO2 atmospheric concentrations were 40 to 50% lower than outdoor concentrations during the spring−summer and autumn−winter periods, respectively. In large cities such as Toronto (Population 2,700,000), the influence of a single major road on outdoor and indoor NO2 concentrations is predominantly masked by spatially-extensive high-density traffic. Machine Learning Using Topology Signatures For Associative Memory
This thesis presents a technique to produce signatures from topologies generated by the Growing Neural Gas algorithm. The generated signatures have the following characteristics: The signature's memory footprint is smaller than the "real object" and it represents a point in the n x m multidimensional space. Signatures can be compared based on Euclidean distance and distances between signatures provide measurements of differences between models. Signatures can be associated with a concept and then be used as a learning step for a classification algorithm. The signatures are normalized and vectorized to be used in a multidimensional space clustering. Although the technique is generic in essence, it was tested by classifying alphabet and numerical handwritten characters and 2D figures obtaining a good accuracy and precision. It can be used for many other purposes related to shapes and abstract typologies classification and associative memory. Future work could incorporate other classifiers.
